Laboratory extruders for pharmaceutical product development

The Caleva Extruder 20 is a screen type extruder, designed to replicate production extrusion on the laboratory bench top.

 

The unique Caleva system of roller extruders is designed to impart very little work to the product as it is processed.  This reduces problems caused by overheating or drying out of the material.

 

Screens of different apertures and heights can be supplied to complement any development program.

 

The diameter of spheroids produced is largely determined by the diameter of the extrudate.

 

The Extruder 20 has a simple operating structure and a small footprint to fit easily on the bench top.


The Extruder 20 is a much improved development from the successful Caleva Extruder 25.  A bench top apparatus suitable for laboratory research and small batch production.

 

The new Extruder 20 has significant advantages over the Extruder 25 that it replaces.

 

The Extruder 20 remains a bench-top machine.

 

The extruder mechanism in this lab scale equipment is identical to production screen extruders.  This will minimize issues with scale-up that can occur when the results of development work is transferred to production.

The product throughput of the Extruder 20 is significantly greater than the Extruder 25.

The smallest batch size that can be extruded is reduced to about 30 grams.  Sample sizes of several kilos present no problem for the Extruder 20.

 

Product recovery (useful extrudate produced as a % of material added to the extruder) is very high and there are few “dead” spaces where material can collect.

 

The extrusion pressure on the extrusion screen is spread more evenly on the Extruder 20. Screens will have a longer effective life.

 

Caleva production extruders use a unique roller extruder mechanism that pushes the material through the screens and does not scrape the material through as with other extruders. This imparts less work on the product and this may be important in sensitive formulations and is therefore particularly suitable for use in the pharma industry.


Output and productivity

The achievable throughput is dependent on the properties of the material being extruded and the screen size used.

 

Although the Extruder 20 is not designed as a production extruder for continuous use, some materials have been extruded at rates equivalent to 25kg per hour.

 

The effective minimum load requirement is about 30 to 50g for a full sized screen.

 

If a reduced height screen is used experiments have shown that if batch sizes of about 10g are used then a recovery of about 7g of extrudate material can be achieved.

Available options

All machines are supplied with a 1mm aperture 150mm diameter extrusion screen.  Additional screens are available as required.

Amongst the options available are:-

·    Screens with apertures from 0.5mm to 3.0 mm can be supplied in 0.1 mm increments as full height, half height and one third height.  Specific requirements should be discussed with us before ordering.

·    All screens and rollers are interchangeable within each height of screen.

·    Replacement screens can be supplied as needed.

A note about terminology:-

Spheronization (or spheronisation), marumerization, pelletization and micropelletization all mean the same thing. The manufacture of products in small regular shaped spheres for use in pharmaceutical and other industries. Generally within each batch the spheroids are not only of regular shape but also of regular size.

The products produced by these processes are called either spheroids, spheres, micro-spheres, pellets and micro-pellets. All these terms refer to the same thing. The size range is normally from about 0.8 mm to about 2.5 mm in diameter.

Granulation normally refers to the manufacture of irregularly shaped particles with a large size range included within the batch.
